Abstract

A dispersion and differential centrifugation technique for representatively sampling microorganisms from soil offers the possibility to deterime microbial bound nitrogen in soil and the C/N ratio in the microbial biomass by a direct method. The method of Hopkins (HOPKINS et al. 1991) was modified and their significance proved to make it useful for the determination of carbon and nitrogen in the separated soil microbial biomass. Using a calibration curve of added bacteria to soil and quartz sand the method was proved. Additionally the new method was compared to the traditional SIR method. First experiments using the modifyed dispersion and differential centrifugation method were done in a field experiment following K15NO3 fertilization (80 kg N ha −1,10% marked).
