Zusammenfassung
Die Ätiologie von Kraniosynostosen ist mit hoher Wahrscheinlichkeit multifaktoriell. Für die syndromalen Kraniosynostosen sind genetische Defekte verantwortlich, die entweder spontan auftreten oder zumeist durch einen autosomal dominanten Erbgang weitergegeben werden. Von dem betroffenen Gendefekt kann jedoch oftmals nicht auf den jeweiligen Phänotyp geschlossen werden, der auch innerhalb einer betroffenen Familie erheblich variieren kann. Neben den genetischen Grundlagen spielen auch zahlreiche andere Faktoren eine Rolle. So können neben infektiösen Ursachen auch metabolische oder hämatologische Erkrankungen sowie exogen zugeführte Teratogene beteiligt sein. Sekundäre frühzeitige Nahtverschlüsse werden durch einen im Säuglingsalter abgeleiteten Hydrozephalus, Enzephalozelen und bestimmte Hirnfehlbildungen wie z. B. die Holoprosenzephalie oder auch durch eine Mikrozephalie begünstigt.
