Mapping of HLA Class II Genes by Pulsed Field Gel Electrophoresis and Size Polymorphism

Abstract

The HLA-D histocompatibility region encodes for polymorphic cell-surface class II antigens composed of a 34 kd α chain and a 28 kd β chain. This region is divided into several subregions:, DP, DO/DZ, DQ and DR, although each contains a complex array of genes and pseudogenes. In this report, the organization of the genes in the HLA-D region of serologically different HLA-homozygous B cell lines was examined by pulsed field gel electrophoresis (1,2) (PFGE) using α and β chain probes from all of the subregions including DVβ (3) (a new HLA class II pseudogene) and a DX region-specific probe. The map was determined to be: DPβ-DPα-DO(DZ)α-DOβ-DXβ-DXα-DVβ-DQβ-DQα-DRβ-DRα. Interestingly, the distances not only between DQα and DRβ but also between DVβ and DQβ were found to vary markedly among different HLA haplotypes.
