Summary
The hydroxylation of aromatic rings by hepatic microsomal drug enzymes was studied by using naphthalene as substrate. Enzyme activity was measured fluorimetrically after extraction into n-butanol by the decrease of the naphthalene concentration. The turnover of naphthalene was greater than that obtained during oxidative demethylation of p-nitroanisole. This coincides with the observation of a smaller Michaelis constant for naphthalene. Naphthalene hydroxylation can be stimulated in a manner similar to the induction of p-nitroanisole O-demethylase by previous treatment of the animals with DDT and phénobarbital. Mouse liver microsomes, however, are not stimulated by DDT.
Zusammenfassung
Die Hydroxylierung aromatischer Ringverbindungen durch mikrosomale Enzymsysteme wurde am Beispiel des Naphthalins untersucht. Als Parameter für die Aktivität wurde die Abnahme der Naphthalinkonzentration nach Ausschütteln in Butanol fluorimetrisch gemessen. Die Umsatzgeschwindigkeit von Naphthalin war höher als die von p-Nitroanisol während seiner oxydativen O-De-methylierung. Dies stimmt mit der Beobachtung einer kleineren Michaeliskonstanten für Naphthalin überein. Die Naphthalinhydroxylierung läßt sich durchVorbehandlung der Tiere mit DDT und Phenobarbital in ähnlicher Weise steigern wie die O-Demethylierung von p-Nitroanisol. Mäusemikrosomen lassen sich durch DDT nicht stimulieren.
