Zusammenfassung

In der Geschichte der Enzymforschung nehmen die Glykosidasen eine ganz besondere Stellung ein: Die spezifische katalytische Wirkung, welche Extrakten von lebenden Zellen zukommt, wurde zuerst an der Hydrolyse von Kohlenhydraten erkannt.

Planche zeigte 1810, daß Extrakte von Pflanzenwurzeln eine Substanz enthalten, die Guajactinktur blau färbt, und Payen und Persoz isolierten 1833 die „Diastase“ aus Gerstenmalz, nachdem schon Kirchhoff (1814) beobachtet hatte, daß eine Verzuckerung der Stärke auch durch einen wäßrigen Malzauszug erfolgt. Zu den ersten Enzympräparaten, die genauer untersucht wurden, gehören das „Emulsin“ aus Mandeln und das „Invertin“ aus Hefe. Der Meinung, daß die Enzyme Proteine sind, hat E. Fischer auf Grund seiner Untersuchungen über Glykosidasen zuerst Ausdruck gegeben1. Auch die ersten Ansätze zum Kernproblem der Enzymologie, der rationellen Erklärung der Spezifität, wurden an Hand der Untersuchungen von glykosidspaltenden Enzymen von E.
