Zusammenfassung
Ribonucleasen greifen Internucleotid-Bindungen von Ribonucleinsäuren an und sind damit eine spezielle Gruppe von Phosphodiesterasen1. Ihre Abgrenzung kann auf Grund verschiedener Kriterien erfolgen1:

1.
nach der Art des Substrates, Ribonucleinsäure oder Desoxyribonucleinsäure, damit wesentlich abhängend von der Art der Pentose und mit entsprechender Rückwirkung auf den Reaktionsmechanismus (Fehlen von 2′-OH in DNS);

2.
nach der Lage der angegriffenen Bindung in der Polynucleotid-Kette als Exonuclease oder Endonuclease (in Analogie zu Exopeptidase bzw. Endopeptidase);

3.
nach der Natur der Spaltprodukte mit 3′-Phosphoryl- oder 5′-Phosphoryl-Endgruppen (was speziell zur weiteren Unterteilung von Ribonucleasen und Desoxyribonucleasen nützlich zu sein scheint);

4.
nach den Strukturerfordernissen in der Umgebung der sensitiven Bindung, z.B. Purin- oder Pyrimidin-Derivate; freies Hydroxyl in 3′- oder 5′-Stellung;

5.
nach der Form des makromolekularen Substrates als einsträngiges oder zweisträngiges Molekül2.
