Zusammenfassung

[3.2.1.30 ß-2-Acetylamino-2-deoxy-D-glucosid acetylaminodeoxyglucohydrolase.] Entdeckung. Erstmals berichteten im Jahre 1929 Karrer u. Mitarb.8 über den enzy-matischen Abbau eines aus Glucosamin aufgebauten Polysaccharides. Sie behandelten feingemahlenes Chitin (aus Hummerschalen) mit dem Saft von Weinbergschnecken (Helix pomatia) und erhielten als Produkt der enzymatischen Chitinhydrolyse N-Acetyl-glucos-amin in kristallisiertem Zustand. Diesem chitinspaltenden Enzym, dessen pH-Optimum bei 5,2 liegt, gaben sie den Namen Chitinase.

Einer Ammoniumsulfat-Fraktionierung wird eine Gel-Filtration über Sephadex G-100 angeschlossen, der erneut eine Präcipitation mit Ammoniumsulfat und eine anschließende Chromatographie über DEAE-Cellulose folgt. Das Enzym spaltet p-Nitrophenyl-2-acetamido-2-desoxy-ß-D-glucopyranosid, setzt aber auch 26% des Glucosamins aus dem ar Glycoprotein des menschlichen Plasmas frei, wenn vorher alle Acyl-neuraminsäure und mindestens 80 % der Galaktose enzymatisch abgespalten worden sind.Google Scholar
