Zusammenfassung
Wie wohl in keinem anderen Teilgebiet der Virusforschung hatte die Anwendung einer besonderen Technik so große Erfolge zu verzeichnen wie bei der Züchtung der Virusarten. Nachdem man sich jahrelang erfolglos bemüht hatte, virusartige Infektionsstoffe auf toten Nährsubstraten, wie sie in der bakteriologischen Züchtungstechnik üblich sind, in vitro zur Vermehrung zu bringen, wurde dieses Ziel durch die Anwendung der von Harrison (1907) inaugurierten und von Burrows, Carrel und A. Fischer ausgebauten Technik der Gewebeexplantation erreicht. Auch die Viruszüchtung in der Gewebekultur hatte allerdings eine bestimmte Entwicklung zu durchlaufen, bis sie zu zuverlässigen und reproduzierbaren Ergebnissen führte. So gelang es zunächst lediglich, das Virus im überlebenden Gewebe zu konservieren oder höchstenfalls in einigen wenigen Kulturpassagen in infektionstüchtiger Form weiterzuführen; der Nachweis aber, daß in Gewebsexplantaten nicht nur eine ausgiebige Vermehrung, sondern auch eine Dauerzüchtung von Virus möglich ist, wurde bekanntlich erst viel später geleistet. Die Hauptschwierigkeit, die sich der Viruszüchtung zunächst entgegenstellte, war zweifellos in der komplizierten und subtilen Technik der Gewebezüchtung begründet, und es ist deshalb kein Zufall, daß die ersten einwandfreien Viruszüchtungen nur von einigen wenigen, mit der Methode der Zellzüchtung völlig vertrauten Forschern erreicht wurden. So war es namentlich Carrel (1925/26), der in einer Serie von meisterhaften Versuchen den strikten Beweis führte, daß sich das Virus des Rousschen Hühnersarkoms in Gewebsexplantaten quantitativ vermehrt und dauernd in Kulturpassagen weiterführen läßt.
