Abstract

Weiler (1956) reported that, coincident with aminoazo dye carcinogenesis, there was loss of organ specificity from rat liver as determined by complement fixation test. Using fluorescein isocyanate-labelled organ specific antisera as histoserological stains, he demonstrated differential staining between rat liver and rat hepatoma. He regarded this lack of staining of tumour cells as confirmatory evidence of loss of their organ specific antigens. — All attempts to produce organ specific antisera by Weiler’s or other methods were unsuccessful and fluorescein isocyanate serum protein conjugates prepared from a wide variety of non-immunized animals gave identical differential staining. The mechanism of this staining will be discussed. — A large series of neoplastic lesions was investigated and it was found that malignant tumours uniformly showed greatly diminished fluorescence compared with the parent tissues. On the other hand innocent tumours and hyperplastic conditions uniformly fluoresced brightly and showed fluorescein-protein affinities similar to those of the tissue of origin. — More recently, other workers have reported results partly at variance with these; however they used fluorescein isothiocyanate instead of fluorescein isocyanate. Miller, Willey, Leestma and Riggs (1963) investigated this problem and found that if fluorescein isocyanate was utilized in preparing the stain they could produce an easily recognizable and consistent difference in cytoplasmic fluorescence between benign and malignant cells but if fluorescein isothiocyanate was used the results were equivocal and inconsistent.
