Zusammenfassung
Nach den Untersuchungen von Willibald Kühne(1877 –1882) über die Bleichung und Regeneration des in den Stäbchen der Wirbeltiere reichlich vor handenen Sehpurpurs glaubte man lange Zeit, der Mechanismus der Dunkel- und Helladaptation bestünde im wesentlichen in einer Zunahme und Abnahme der Photopigmentkonzentration in den Sinneszellen der Netzhaut. Zweifel an einer solchen photochemischen Theorie der Helligkeitsanpassung wurden bereits frühzeitig geäußert. Fritjof Holmgren, einer der Entdecker des Elektroretinogramms, beobachtete 1882 beim Kaninchen auch nach vollständiger Bleichung des Sehpurpurs auf Lichteinfall Schwankungen des Retinastromes, woraus er folgerte, daß die Erregung der Netzhaut in keiner wesentlichen Beziehung zu den Bleichungsund Regenerationserscheinungen des Sehpurpurs stehe. Zu ähnlichen Schlußfolgerungen gelangten Granit, Munsterhjelmund Zewi(1939), die bei Fröschen und Katzen während Dunkeladaptation keine direkte Beziehung zwischen der Amplitude der b-Welle im Elektroretinogramm und der gleichzeitig gemessenen Sehpurpurkonzentration feststellen konnten. Weitere experimentelle Einwände gegen die photochemische Theorie der Helligkeitsanpassung wurden von Baumgardt(1950) und Haginsund Rushton(1953) vorgebracht, wonach die Reizschwelle eines dunkeladaptierten Auges bis zum 106fachen ansteigen kann, ohne daß nennenswerte Bleichung des Sehpurpurs eintritt.
