Zusammenfassung
Histamin ist eine kleinmolekulare, in Wasser, Alkohol und heißem Chloroform gut lösliche Base, hingegen ist seine Löslichkeit in wasserfreiem Äther nur gering. Ein ähnliches Lösungsverhalten zeigt auch sein Chlorhydrat, jedoch löst sich dieses kaum noch in heißem Chloroform. Gegen Erhitzen bei saurer Reaktion ist Histamin stabil und wird selbst durch kurzes Kochen mit 3 n-Salzsäure nicht zerstört. Wie alle Amine bildet es mit zahlreichen Alkaloidfällungsmitteln schwer-lösliche Komplexsalze. Diese wichtigen Eigenschaften des Histamins sind die Grundlage aller Isolierungs-Methoden. Der erste Schritt jeder Reinigung ist die Abtrennung von Begleitstoffen. Schwierigkeiten treten dabei auf, wenn die Base aus fett- und eiweißreichen Organhomogenaten, aus Blut, Urin oder aus pflanzlichem Gewebebrei zu isolieren ist. Selten wird es dann gelingen, in ein oder zwei Reinigungsstufen das für biologische und biochemische Untersuchungen oder für eine sich anschließende Kristallisation geeignete Rohprodukt zu erhalten, vielmehr wird man durch eine sinnvolle Kombination von verschiedenen Fällungsmethoden mit Extraktionen oder Dialysen von Fall zu Fall einen geeigneten Weg für jedes spezielle Rohmaterial zu suchen haben. In diesem Abschnitt wird anhand von erprobten Beispielen demUntersuchenden gesagt werden, welche Möglichkeiten einer Isolierung von Histamin aus biologischem Material sich bieten, jedoch läßt sich der Wunsch nach einer allgemein gültigen Arbeitsvorschrift naturgemäß nicht erfüllen.
