The influence of α-tocopherol-nicotinate (Renascin®) on cardiac glycoside effects

Summary

The influence of α-tocopherol-nicotinate and related compounds on cardiac glycoside effects on isolated guinea-pig atria and on the glycoside binding to guinea-pig heart homogenate was investigated.

α-tocopherol-nicotinate and dodecanoic acid had an inhibitory effect on the positive inotropic action of digoxin but not on that of ouabain. This effect was parallel to their inhibitory effect of digoxin and not ouabain binding to heart homogenate. The tested compounds showed a protective effect against cardiac glycoside induced arrhythmias which is not in correlation with their effects on the positive inotropic action of the glycosides or on their binding to heart homogenate. The protective effect may be due to a direct action of the drugs on the cell membrane.
