Zusammenfassung
Defektproteinämien wie bei α1-Antitrypsin-Mangel weisen oft erstaunliche Entdeckungsgeschichten auf. Der Zusammenhang zwischen α1-Antitrypsin-Mangel und dem Lungenemphysem wurde erst in den 60er-Jahren erkannt, und danach konnten die pathophysiologischen Zusammenhänge zwischen Proteaseinhibitoren und Destruktion der elastischen Lungenkräfte (Rebmann 1989; Carrell u. Lomas 2002) sowie die Bedeutung der Infektionen und die Rolle der Granulozyten entschlüsselt werden. Hervorzuheben ist die Bedeutung der komplexen Interaktion zwischen molekularen genetischen Grundlagen und Umweltfaktoren (Rauchen).
