Gewebethromboplastin: Biochemie, Molekularbiologie und Physiologie

Zusammenfassung

Gewebethromboplastin wurde zuerst als die gerinnungsaktivierende Substanz in Gewebeextrakten beschrieben. Da die Extraktion von Lipiden die Aktivität von Gewebethromboplastin zerstörte, wurde ein Gerinnungsfaktor bestehend aus einem Lipid- und einem Proteinanteil postuliert. Die Isolierung des Proteinanteils hat sich als extrem schwierig herausgestellt, wie bei vielen anderen zellständigen Molekülen. Die cDNA für Gewebethromboplastin wurde 1987 von mehreren Arbeitsgruppen Moniert. Gewebethromboplastin wird heute in der englischsprachigen Literatur als Tissue factor (TF) bezeichnet. Scit der Klonierung von TF sind die Prinzipien der Funktion, die molekulare Struktur und die Regulation des Gens auf der zellbiologischen Ebene weitgehend aufgeklärt worden. Die Inaktivierung des TF-Gens in der Maus und Befunde aus der Tumorbiologie legen nahe, daß TF sowohl das extrazelluläre Milieu durch die Aktivierung der Gerinnung verändern kann als auch die Zellfunktion durch Signalübertragung wesentlich beein-flußt. Die zytoplasmatische Domäne von TF bindet spezifisch an „actin-binding protein 280“! (ABP-280), und diese Interaktion ist potentiell ; für die zellbiologischen Effekte von TF von Bedeutung. In den nächsten Jahren werden diese j neueren Funktionen des Gebethromboplastins in der Angiogenese, Tumorzellmetastasierung und -invasion sicherlich im Detail aufgeklärt werden.
