Immunologisches Monitoring von Patienten mit allogenen Knochentransplantaten

Summary

Allogeneic bone grafts are necessary for the reconstruction of destroyed skeletal parts. The immunological response of the recipient against these allogeneic grafts is still controversal. The recipients of allogeneic bone grafts were followed up by immunological methods like MLC, flow cytometry, and MAILA. Employing this set of different tests, we succeeded in detecting class I and class II antigens on the surface of bone cells. There was a strong correlation between the immunological mismatch between recipient and donor and the degree of the immunoreaction.

Zusammenfassung

Bei der Rekonstruktion morphologisch veränderter und funktionell gestörter Abschnitte des Bewegungsapparates kann derzeit auf den Einsatz von allogenem Spenderknochen nicht völlig verzichtet werden. Das immunologische Schicksal von allogen transplantiertem Knochen ist dabei weitgehend noch ungeklärt. Mittels MLC, DurchfluBzytometrie und MAILA werden die allogenen Knochenempfänger immunologisch weiterverfolgt. Auf diese Weise gelingt es, spezifische Antikörper des Empfängers gegen HLA-Klasse-I und -II-Antigene des Spendergewebes nachzuweisen. Dabei findet sich eine negative Korrelation zwischen dem Ausmaß der Immunreaktion im Sinne einer Antikörperstimulation und dem Grad der HLA-Übereinstimmung.
