X-ray and Neutron Scattering Studies of Lipid Monolayers and Single Bilayers

Abstract

To understand the behavior of self assembled macromolecular arrays of mole-cules found in biological materials, one must know the structure of these as-semblies. Two of the fundamental techniques used for obtaining molecular ar-rangements with Å resolution are x-ray and neutron scattering. Studies of the structure of bulk, polycrystalline samples of lipids give a wealth of information about the behavior of multibilayer systems such as the bilayer repeat spacing, molecular tilt angle, chain packing area, and headgroup hydration. However, it is not clear how many multilayer properties are shared by the same single bilayer. For example, do the various phases found in multi-lamellar arrays exist in single, free bilayers. In the case of these ultra-thin organic structures, standard diffraction techniques are not applicable because of the small, scattered signal and substantial background contribution. However, x-ray and neutron surface techniques can provide a means for this needed structural characterization.
