Abstract

Bifidobacterium represents one of the most numerous bacterial genera in the gut of humans and other animals and plays a fundamental role in the health of the host. In view of the development of bifidobacteria probiotic strains with improved characteristics, there is considerable interest in their manipulation using genetic engineering techniques. In spite of their importance, very little is known about the genetics of these microorganisms. Application of recombinant DNA technology to bifidobacteria was delayed by past difficulties associated with the transformation of members of this genus. Only recently has the successful transformation of Bifidobacterium by electroporation been reported (Missich et al. 1994; Argnani et al. 1996, Rossi et al. 1997; Matsumura et al. 1997). An optimized procedure for the electroporation of plasmid DNA into several Bifidobacterium species is described.
