Genetic Diversity of Ralstonia solanacearum in Japan
Five races and 5 biovars of Ralstonia solanacearum have been described and they differ in host range, pathogenicity and physiological properties. In Japan, R. solanacearum was isolated from various hosts, and 2 races and 4 biovars were identified. RFLP and PCR-RFLP analysis of the 16S and/or 23S ribosomal DNA region showed that the Japanese race 1/biovars 1 – 4 strains were similar to Asian biovars 3, 4 and 5. Japanese race 3/biovar 2 strains were very similar to strains of American biovar 1 and Blood disease bacteria (BDB) from banana. RFLP and rep-PCR analyses of Japanese strains differing in geographical origin and host resulted in two major clusters. Cluster 1 comprised Japanese race 1/biovars 1 – 4. Cluster 2 comprised Japanese race 3/biovar 2. Cluster 1 was further subdivided into 7 groups by RFLP analysis and 4 groups by rep-PCR analysis, respectively. Clusters correlated with a race, biovar, tobacco compatibility and original host.
