Abstract

Since its introduction by Faulk and Taylor in 1971, the immunogold staining method has undergone an enormous evolution. Methods were developed to prepare gold sols in a wide range of sizes, with a narrow size distribution enabling double or even triple labeling in electron microscopy (EM). A large variety of macromolecules other than antibodies (e.g., streptavidin, protein A, lectins, enzymes, and ligands such as insulin) were coupled to colloidal gold particles. Gold probes have not only proven to be efficient markers for EM, but in combination with silver enhancement, their use has been extended to the light microscopic and macroscopic level (immunoblotting).
