Die alkalische Leukocytenphosphatase hydrolysiert das Substrat α-Naphthylphosphat; das dabei frei werdende Naphthol wird mit einem Diazonium-salz gekoppelt, mit dem es einen unlöslichen Azofarbstoff bildet.
