Summary
It is shown that the effects of various synthetic gestagens on the uterus and the pituitary gland may differ considerably. Some possible explanations are:
-
1.
The interference of other activities i.e. oestrogenicity with progestational effects.
-
2.
Physiological differences in various animal species such as the relative importance of ovarian and placental function for the maintenance of pregnancy.
-
3.
The presence or absence of central effects.
-
4.
Properties of receptors.
Metabolic differences between some derivatives of pregnane and retropregnane are discussed.
The examples mentioned show that in a number of instances molecular biological studies can yield an explanation for the different effects observed with related substances in the same animal species, and for those of one substance in various species. More rarely the analyses of metabolites will serve this purpose. It is hardly to be expected that this kind of study will allow to predict the presence of desired or undesired effects of new compounds.
