Effects of Ethanol on Gluconeogenesis in the Perfused Rat Liver

Abstract

My interest in the biochemistry of alcohol arose recently from observations made in experiments on the rate of gluconeogenesis in the perfused liver in the presence of lactate and glycerol. On simultaneous addition of glycerol and lactate the rates of gluceneogenesis were found to be higher than the sum of the rates found with the two substrates alone. Thus, lactate alone formed glucose at a rate (umole/min/g) of 1.06, glycerol of 0.48 and both together of 1.90. As glycerol, in the form of α-glycerophosphate, can act as a hydrogen donor in the cytoplasm we thought that the enhanced rate of gluconeogenesis might be connected with the reducing capacity of glycerol.
