Zusammenfassung

Sehr bald nach der Entdeckung des hohen Jodgehaltes der Schilddrüse durch Baumann (1896) hatte sich die Erkenntnis herausgeschält, daß das Thyreoglobulin im Kolloid der Schilddrüsenfollikel das Depot für das Hormon der Schilddrüse darstelle, und daß sein Jodgehalt gewissermaßen ein Index für seine Wirksamkeit bedeute. So wird auch heute noch in der USA-Pharmakopoe der Jodgehalt eines Schilddrüsentrockenpulvers als Maßstab für seine Reinheit angegeben; er muß mindestens 0,2% betragen, um als ausreichend wirksam akzeptiert zu werden. Oswald gelang es um 1910, aus dem Thyreoglobulin das Dijodtyrosin zu isolieren, während Kendall im Jahre 1916 die kristallinische Isolierung des Thyroxins durchführte. Aber erst als Harington (1927) das Thyroxin in seiner chemischen Struktur aufgeklärt hatte und ihm die Synthese des Thyroxins gelungen war, begann man in der Klinik exakte Werte für die Hormoneffekte in Relation zur Dosis zu gewinnen. Damals blieb es unerklärlich, daß Thyreoglobulin einen wesentlich stärkeren Hormoneffekt erreichte, als es seinem Thyroxingehalt entsprach — wobei sich auch hier die Ausgangslage des Grundumsatzes entsprechend dem Wilderschen Ausgangsgesetz als wesentlich erwies (Thompson C. S. 1935) —, so daß andere jodierte hormonwirksame Eiweißverbindungen postuliert wurden. Auch war es auffallend, daß Thyroxin nur in alkalischem Milieu wirksam wurde, weswegen besonders Thompson bei seinen systematischen Studien über den den Basalstoffwechsel steigernden Effekt des Thyroxins und anderer ihm ähnlicher Verbindungen die Notwendigkeit der Verwendung alkalischer Zusätze unterstrich (1934).
