The spleen shows a high content of hydrolases. Lysosomal enzymes as well as esterase predominate in the reticulohistiocytic cells of the red pulp. Membrane enzymes such as alkaline phosphatase and nucleoside phosphatases predominate in the white pulp. The well known morphologic and functional species differences of the spleen are reflected in different enzyme patterns.

Various oxydoreductases are histochemically demonstrable in the spleen.

Following radiation damage to the spleen, pronounced increases in lysosomal enzymes and esterases occur in the radiation-resistant reticulohistiocytic cells during the phases of destruction and resorption.

A scarcity of enzymes is evident in the still undifferentiated islands of regeneration during the recovery phase.
