Summary
The following data on the regulatory mechanism of the human spleen in the turnover of thrombocytes seem to be significant. They were obtained from comparative studies on platelet kinetics, surface activity measurements, catecholamine stimulation before and after splenectomy, and studies of cases with normal spleen and with hypersplenism:

Normally 1/3 of platelets are pooled in the spleen. There is an exchange mechanism between this pool and the circulating thrombocytes (normal platelet pool).

In hypersplenism the splenic pool for platelets ranges between 40 and 90% of the total number of thrombocytes (increased splenic pool). This results in a significant thrombocytopenia with a slightly shortened life span of platelets.

After a life span of 8 to 11 days platelets are destroyed predominantly in the spleen.
