Platelet Aggregability, Disaggregability, and Serotonin Uptake in Migraine

Changes discovered in serotonin (5-HT) metabolism during migraine attacks have focused attention on blood platelets, the main reservoir of 5-HT in the body. Several disturbances in platelet function were indeed observed in migraineurs, and two lines of thought emerged. In one, the platelet abnormality was thought to result from a circulating agent which causes 5-HT release. The activity of this agent was thought to be directly connected to the initiation of the migraine attack itself (e.g., through the release of other vasoactive substances), or the released 5-HT could act as an intermediate. This view was first suggested by Anthony and co-workers [1, 2]. We have shown that a plasmatic factor occurs during migraine attack which can initiate 5-HT release from platelets of normal subjects. The responses of migraineurs’ platelets were quite similar to those of controls [3].
