Abstract

Clinical trials with new chemotherapeutic agents must be preceded by extensive pharmacologic study in animals. Freireich [1] reviewing a large number of experimental data showed that there is a close relationship between the LD 10 in small animals and the maximum tolerated dose (MTD) in man, when the values are given on a mg/m2 basis. The ratio of these values is always very close to 1, even when the doses for the different drugs vary with a ratio of 2,000 to 1 (Fig. 1).
