Abstract

The first three steps of ether-lipid biosynthesis are catalysed by enzymes located in the peroxisomal membrane, whereas the further steps are associated with the endoplasmic reticulum [4,9]. The peroxisomal enzymes are dihydroxyacetonephosphate (DHAP) acyltransferase, alkyl-DHAP synthase and alkyl-DHAP reductase. Dihydroxyacetonephosphate acyltransferase (DHAP-AT, E.C. 2.3.1.42) is found only in peroxisomes [3] and, therefore, can be used as an enzymatic marker of this organelle [8]. DHAP-AT’s active site is on the inner face of the membrane. The enzyme catalyses the first step in the synthesis of ether lipids, which have acyl-DHAP as an obligate precursor.
