Zusammenfassung
Das Ziel der hier vorgestellten Untersuchung war es, zu prüfen, ob Pentaerithrityltetranitrat (PETN) in der Lage ist, funktionelle und morphologische Änderungen im kardiovaskulären System zu beeinflussen, die durch eine Langzeit-NO-Synthase-Hemmung provoziert werden können. Die für die Untersuchung verwendeten 10 Wochen alten Wistar-Ratten wurden in 3 Gruppen geteilt: Die erste Gruppe diente als Kontrolle. Die zweite Gruppe wurde mit in Trinkwasser gelöstem (50 mg/Kg) NG-Nitro L-Argininmethylester (L-NAME) behandelt. Die dritte Gruppe wurde behandelt mit L-NAME (50 mg/Kg) in Trinkwasser plus PETN (2×50 mg/Kg). Das Experiment dauerte 6 Wochen. Blutdruck wurde indirekt mit Hilfe der Plethysmographie an der Schwanzarterie gemessen. Für die funktionellen Untersuchungen wurden isolierte Ringpräparationen der thorakalen Aorta benutzt. Diese wurden suspendiert in einem Organbad, welches modifizierte Krebslösung enthielt. Sie waren verbunden mit einem Kraftaufnehmer zur Messung der isometrischen Spannung. Für die morphologischen Untersuchungen wurden die Tiere getötet. Sie wurden dann zur Fixierung über den linken Ventrikel mit Glutaraldehyd perfundiert. Der Perfusionsdruck wurde auf 120 mmHg fixiert. Die thorakale Aorta (TA), die Arteria carotis (CA), der septale Schenkel der linken absteigenden Koronararterie (RS) wurden entnommen und für die elektronenmikroskopische Untersuchung aufgearbeitet. Die Wanddicke (WT, wall thickness) und der innere Durchmesser (ID) wurde unter einem Lichtmikroskop ausgemessen und die WT/ID-Ratio errechnet. In der Gruppe von Ratten, die mit L-NAME+PEIN behandelt waren, war der systolische Blutdruck (163 ± 1 mmHg) signifikant niedriger als in der L-NAME behandelten Gruppe (172 ± 2 mmHg). Allerdings war er auch in dieser Gruppe immer noch höher als in der Kontrollgruppe (126 ± 2 mmHg). 6 Wochen nach Behandlung mit L-NAME war die endothelabhängige Acetylcholin-induzierte Relaxation der isolierten thorakalen Aorta signifikant vermindert. Der inhibitorische Effekt von L-NAME konnte durch die gleichzeitige Behandlung mit PETN aufgehoben werden. In allen Arterienpräparationen provozierte die NO-Synthase-Hemmung einen deutlichen Anstieg von WT, CSA und WT/ ID. Dieser Effekt war außer in RS signifikant. ID nahm lediglich in TA zu. Die Langzeitgabe von L-NAME zusammen mit PETN verhinderte die Zunahme von WT, CSA und WT/ID in allen untersuchten Arterien mit Ausnahme von TA. ID war ausschließlich in TA angestiegen. Die hier vorgestellten Daten belegen, dass funktionale Störungen und strukturelle Veränderungen im kardiovaskulären System, wie sie durch eine Langzeit-NO-Synthase-Inhibition proviziert werden können, durch eine gleichzeitige Gabe des exogenen NO-Donators PETN abgeschwächt werden können.
