Zusammenfassung
Es wird eine empfindliche und selektive Routinemethode zur gleichzeitigen Bestimmung von Pentaerithrityldinitrat (PEDN) und Pentaerithritylmononitrat (PEMN) in Humanplasma unter Verwendung von Isosorbid-5-mononitrat (IS-5N) als internem Standard beschrieben. Die Analyte wurden durch Extraktion mit Hexan/Essigester (1:4, v/v) aus Humanplasma isoliert und mit N,O-Bis(trimethylsilyl)trifluoracetamid (BSTFA) zum Trimethylsilylether (TMS-ether) derivatisiert. Die Quantifizierung erfolgte nach gaschromatographischer Auftrennung massenspektrometrisch nach negativ chemischer Ionisierung (GC-NICI-MS) unter Verwendung von Methan als Reaktandgas. Alle Analyte wurden bei der Masse m/z 46=[NO2]− quantifiziert. Die Bestimmungsgrenze lag bei 1 ng/ml für PEMN und bei 0,2 ng/ml für PEDN. Lineare Eichgeraden wurden über den Konzentrationsbereich 1–150 ng (PEMN) und 0,2–30 ng (PEDN) erhalten. Für die Richtigkeit (accuracy) der Methode wurde ein maximaler Wert von 10,0% (PEDN) bzw. von 9,0% (PEMN), für die Wiederholbarkeit (precision) von 22,2% (PEDN) bzw. von 16,3% (PEMN) berechnet.
Die beschriebene analytische Methode, die nach den GLP-Richtlinien validiert wurde, erlaubt die zuverlässige Quantifizierung von PEDN und PEMN in Plasmaproben aus pharmakokinetischen und klinischen Studien. Die Bioanalytik kann somit das biomedizinische Forschungsprogramm zu Pentaerithrityltetranitrat (PETN) ⋆ wirkungsvoll begleiten.
