Zusammenfassung

Dieses Übersichtsreferat erläutert den gegenwärtigen wissenschaftlichen Erkenntnisstand über die Bedeutung von freien Radikalen und reaktiven Sauerstoffspezies (reaktive Oxidantien) in der molekularen Dermatopathologie und Dermatotherapie. Schon die Dermatologen Unna und Gans beschäftigten sich mit der Wechselwirkung von Sauerstoff und Haut. Das Integument ist aufgrund seiner Barrierefunktion neben dem Auge und der Lunge ein Zielorgan des oxidativen Stresses. Wir erläutern den pathophysiologischen Stellenwert reaktiver Oxidantien am Beispiel physiko-chemischer Hautnoxen, Hautentzündung, -ischämie und Karzinogenese. Auf pharmakotherapeutische und dermatotoxikologische Aspekte von Oxidantien und Antioxidantien wird hingewiesen. Das Konzept des oxidativen Stresses als eine signifikante Determinante in der molekularen Dermatopathologie ermöglicht neue experimentelle und klinische Ansätze sowohl in Betrachtungen über die Pathogenese als auch für die Therapie von Hauterkrankungen.

Summary

We present a state of the art-review of the involvement of reactive oxygen species and free radicals (reactive oxidants) in molecular dermatopathology and dermatotherapy. At the beginning of this century, the dermatologists Unna and Gans already studied the interaction of oxygen and skin. Due to its barrier function and content of peroxidizable lipids, skin is a target organ of oxidative injury. The lung and the eye are other examples of oxygen target organs. Oxidative injury is a significant determinant in molecular dermatopathology. We analyze the participation of reactive oxidants in basic dermatopathologic events such as skin irritation, — inflammation, — ischemia and — carcinogenesis. Finally, specific aspects of oxidants and antioxidants in dermatopharmacology and -toxicology are investigated. The concept of oxidative injury in dermatopathology is a useful biomedical simplification and will help to improve our understanding of skin diseases. With the rapid advance occurring in this field of dermatology research, new theoretical and therapeutic approaches may evolve and improve clinical patient management.
