Zusammenfassung
Die chemische Blockade endokriner Drüsen hat nicht nur im Rahmen der Grundlagenforschung, sondern auch in Diagnostik und Therapie an Bedeutung gewonnen. Im Vergleich zur operativen oder durch ionisierende Strahlen induzierten Ausschaltung bieten Endokrinostatica die Möglichkeit der Hemmung von Partialfunktionen. Als Beispiel seien Alloxan und IPTD als Inhibitoren von Inselteilfunktionen und die Thyreostatica vom Thiouracil-, Thiocyanat- und Perchlorattyp genannt. Zu den bekanntesten direkt adrenostatisch wirkenden Substanzen gehören: Amphenon B (3,3-bis(p-aminophenyl)2-butanon), sein Abkömmling SU4885 (2-methyl-l,2-bis(3-pyridyl)-1-propanon), Δ 4-Cholestenon, DDD (2,2-bis(p-Chlorphenyl)-1,1-dichloräthan) und dessen Diäthylderivat Perthan (2,2-bis(p-Äthylphenyl)-1,1-dichloräthan) (ausführliche Literatur bei 1). Den steroidalen Spirolactonen (SC 9420 = 3-(3-oxo-7α-acetylthio-17,3-hydroxy-4-androsten-l7α-yl) propionsäure γ-lacton) können dagegen nur indirekte adrenostatische Eigenschaften zuerkannt werden. Als Aldosteronantagonisten hemmen sie kompetitiv die Mineralocorticoidwirkung an distalen Abschnitten des Nephrons, beeinflussen aber weder Produktion noch Sekretion von Aldosteron.
