Zusammenfassung
Für die quantitative Auswertung von elektronenmikroskopischen Aufnahmen ist eine Eichung der Vergrößerung unerläßlich. Für den Routinebetrieb genügen die von der Lieferfirma angegebenen Vergrößerungen. Die Schwierigkeiten bestehen darin, daß man gewöhnlich an lichtoptisch oder interferometrisch vermessene Objekte anschließt. Bei den höchsten Vergrößerungen bis zu 100000fach werden diese im Endbild größer als der Bildausschnitt abgebildet, so daß bei geringen Vergrößerungen an kleinere Objekteinzelheiten anzuschließen ist. Hierdurch werden die elektronenmikroskopischen Vergrößerungsangaben nie genauer als ±2– 5%. Eine Genauigkeit von ± 1,5 % ist nur unter sorgfältig einzuhaltenden Bedingungen möglich (s. u.).
