Zusammenfassung

Um Bakterienkulturen nach dem Impfen des Nährbodens (Agar) elektronenmikroskopisch zu untersuchen, gibt es folgende Möglichkeiten:

  1. 1.

    Der Nährstoffboden wird direkt mit verdünnter Kollodiumlösung in Amylazetat übergossen (Wyckoff, 1948b). Der getrocknete Kollodiumfilm wird dann mit den haftenden Bakterien auf Wasser abflottiert und auf Trägernetze gefischt. Für dieses und die folgenden Verfahren werden am zweckmäßigsten Glasobjektträger verwendet, die mit einer Agarschicht überzogen werden.

    Mit Kollodium oder Formvar befilmte Glasplatten werden gegen die Kolonie gepreßt, wobei die Bakterien z. T. hängen bleiben (Abklatschverf ahren). Nach Liebermeister (1953) kann man nach dem Auflegen eines bewachsenen Agarblockes auf eine befilmte Glasplatte auch eine Fixation mit Bouinscher Lösung, die auch sonst in der Agarfixation üblich ist, durchführen. Das Agarstück wird dann vorsichtig abgeschleudert. Die anschließende Wässerung erfolgt durch Schwimmen des Präparates auf Wasser, wobei die Schichtseite nach unten zeigt. Man kann auch gleich den befilmten Agarblock auf die Glasplatte legen und die Kultur zwischen Agarblock und Film wachsen lassen.
