Zusammenfassung
Das Griseofulvin ist keineswegs ein „neues Antibioticum“, sondern eine seit 22 Jahren bekannte Substanz mit einer interessanten Geschichte. Wie erinnerlich, wurde das Griseofulvin erstmalig von Oxford, Raistrick und Simonart (1939) aus dem Penicillium griseofulvum Dierckx isoliert. Damals fand es, als man vorwiegend antibakterielle Wirkstoffe suchte, wenig Beachtung. Zum zweiten Male wurde das Griseofulvin durch Brian, Curtis und Hemming (1946) aus Kulturen von Penicillium Janczewski isoliert, allerdings in Form seines biologischen Wirkungsprinzips, des sog. „Curling-Faktors“; das ist ein Effekt, der in einer lockenähnlich verwindenden, bischofsstabartigen Einrollung der Hyphen und Keimschläuche besteht. Ein weiteres Jahr später konnten Grove und McGowan (1947) die Identität von Curling-Faktor und dem bereits 1939 beschriebenen Griseofulvin nachweisen. Die chemische Struktur wurde von Grove U. Mitarb. (1951; 1952) bestimmt. Es hat die Summenformel C17H17O6C1, besitzt also weder eine Hydroxyl- noch eine Carbonylgruppe. Griseofulvin ist stark rechtsdrehend, thermostabil und autoklavierbar, hat einen Schmelzpunkt zwischen 218 und 220° C, läßt sich bei dieser Temperatur leicht durch Sublimation reinigen; es ist schlecht löslich in Wasser (etwa 1 mg-%), besser in organischen Lösungsmitteln (zwischen 1 und 12%).
