Cyclic AMP and Chemotaxis of Leukocytes

Cyclic AMP, Cell Growth, and the Immune Response

Abstract

We wish to report here on the possible role of cAMP in chemotaxis of leukocytes (leukotaxis). Chemotaxis of leukoytes is a cell-stimulatory process by which chemical changes in the environment are recognized. As a consequence of this cellular recognition process, the cell changes its migratory behavior from random to directional locomotion along a positive concentration gradient (Wissler et al., 1972a; Sorkin et al., 1970) without changing the speed of migration (Carruthers, 1966, 1967; Ramsey 1972a, b). Chemotactic responses are shown by phagocytes— neutrophils, eosinophils, and macrophages (for review see Sorkin et al., 1970). Recently, it has also been reported that basophils (Kay and Austen, 1972) and lymphocytes (Ward et al., 1971) give a chemotactic response. It is likely that other tissue cells, like mast cells, histiocytes, etc., show chemotaxis, but the lack of suitable assay systems restricts our knowledge of their chemotactic response. In view of the possible fundamental importance of tactic reactions (chemotaxis, osmotaxis, thermotaxis, phototaxis, aerotaxis) for the development, differentiation, organization, and behavior of living cells, we would like to regard leukotaxis as a model system for many kinds of recognition phenomena.
