Zusammenfassung
Körpereigene Verbindungen werden durch Fluorierung sterisch minimal, chemisch-biologisch jedoch grundlegend verändert. Der Organismus läßt sich durch die sterische Ähnlichkeit täuschen und akzeptiert das Fluor-Analogon, wodurch dieses als Antimetabolit in die Lebensvorgänge eingreifen kann. Bekannt ist die außerordentliche Giftigkeit der Fluoressigsäure, die an Stelle der Essigsäure in den Fettsäure-Cyclus eintritt und diesen blockiert. Nach dem Prinzip „Antimetabolit-Bildung durch Fluorierung“ wurde 1957 von Heidelberger [5] und Chemikern der amerikanischen Hoffmann-La Roche-Werke das 5-Fluoruracil entwickelt, das als verhältnismäßig selektiver Tumorinhibitor Eingang in die Chemotherapie des Krebses gefunden hat. Das 5-Fluoruracil besetzt in der Zelle dem Uracil zustehende Plätze am RNS-Strang, unterbindet die Synthese des Thymins und damit letztlich die Weiterteilung des Zellkerns.
