Renale und intestinale Glucuronidierung von Lithocholsäure

Zusammenfassung

Mikrosomen des Dünndarmes, der Niere und der Leber von männlichen Wistarratten glucuronidieren Lithocholsäure. Die Vmax der LC-UDP-Glucuronyltransferase betrug bei den Kontrollmikrosomen des Dünndarmes 1091 ± 159, der Niere 1271 ± 193 und der Leber 1251 ± 212 pmol × mg Protein-1 × min-1. Nach Vorbehandlung mit Phenobarbital stieg die Aktivität der intestinalen LC-UDP-Glucuronyltransferase um 45,1 %, der renalen um 24,5% und der hepatischen LC-UDP-Glucuronyltransferase um 112,3%. Der KM-Wert betrug für die intestinale LC-UDP-Glucuronyltransferase im Mittel 0,037 mM, für die renale im Mittel 0,032 mM und für die hepatische LC-UDP-Glucuronyltransferase im Mittel 0,040 mM. Vorbehandlung mit Phenobarbital beeinflulßte die KM-Werte nicht. Die extrahepatische Glucuronidierung von Lithocholsäure kann bei Lebererkrankungen von Bedeutung sein.
