Zusammenfassung
Die exakte Kenntnis der molekularen Struktur von Plasmamembranen erlaubt erst eine Zuordnung von Funktionen. Die technischen Probleme einer Reindarstellung von Plasmamembranen sind gelöst. Die Ultrastruktur ist bekannt. Chemisch bestehen Plasmamembranen aus drei Lipoproteinen sowie Glykoproteinen, von denen ein funktionell bedeutsames Glykoprotein bisher am besten charakterisiert ist. Als Markerenzyme sind vor allem die 5 Nukleotidase sowie Kalium-, Natrium- und Magnesium-abhängigen ATPasen, eine membranspezifische Esterase sowie Aminopeptidase zu nennen. Es gibt Hinweise dafür, daß die Basis Lipoproteine, vor allem ein Lipoprotein A mit jeweils unterschiedlicher Enzymaktivität ausgestattet sind und spezifische Funktionen als Rezeptoren bzw. für den Stoff transport besitzen. Für Asialoglykoproteine sowie für einige organische Anionen sind Rezeptoren bekannt bzw. bereits charakterisiert. Über Hormonrezeptoren ist das Wissen noch lückenhaft. Für funktioneile Studien an isolierten Leberzellen sind enzymfrei präparierte Zellen das bessere und den in vivo-Bedingungen adäquatere Ausgangssubstrat. In pathologische Zustände der Leber sind Plasmamembranen vielfältig und z. T. in kritischer Weise verwickelt. Änderungen der Membrankomposition sind bei funktionellen Studien unter pathologischen Bedingungen zu berücksichtigen. Die Bedeutung der Leberzelle als Pinozytose- und Sequestrationsorgan für Immunogene ist durch Änderungen der intrahepatischen Zirkulation klinisch sichtbar geworden. Die physiologische Funktion der Leber für die Elimination von portal anflutenden Antigenen ist noch nicht hinreichend erforscht.
