Abstract
The renin-angiotensin-system is a major regulatory system of blood pressure and cardiovascular function. It has been implicated in the pathogenesis of hypertension in humans as well as in the development of cardiac and vascular hypertrophy. However, studies on the regulation of the genes of the renin-angiotensin-system and their impact on hypertension in humans are difficult to perform. We, therefore, established transgenic rats carrying the human renin or the human angiotensinogen genes. These rats showed tissue-specific expression of the transgenes and secreted the human proteins into the plasma. Due to the species-specificity of the human renin substrate reaction, human renin and angiotensinogen do not interact with the components of the renin-angiotensin-system of the rat. However, in transgenic rats carrying the human angiotensinogen, infusion of human renin elicits a hypertensive response, which can be blocked by a human renin specific inhibitor, Ro 42-5892. The “transfer” of the human renin substrate reaction to rats allows to test human specific enzyme kinetics in the rat, which are suitable models for cardiovascular research. These rats may therefore be test models for the development of human specific renin inhibitors and for studies on the interaction of the human RAS components in the pathogenesis of hypertension. These rats have been considered for patenting.
