Anti-Platelet Treatment of Metastasis

Abstract

Metastasis is a multistep process involving detachment of tumour cells from the primary site, dissemination through the blood stream, arrest in capillary beds, adherence to the vascular wall, dissolution of the basal lamina, migration and growth in extravascular spaces (see Poste and Fidler 1980, Terranova et al 1986, for reviews). The involvement of platelets in the metastatic process has been suggested on the basis of clinical and experimental data (see Gasic 1984, Mehta 1984, Jamieson et al 1987, Weiss et al 1989 for reviews). In the present paper, we will discuss the possible in vitro and in vivo interactions between platelets, tumour cells, endothelial cells and sub-endothelial matrix, the relevance of these multiple interactions for the pathogenesis of metastases and their possible pharmacological modulation for a selective anti-metastatic therapy.
