Interaction between ANP and the Sympathetic Activity

Summary

Three types of mutual interactions between ANP and the sympathetic activity have been identified: 1) Adrenergic atrial stimulation enhances regulated atrial ANP release; 2) the hemodynamic effects of exogenous ANP are blunted by reflex activation of sympathetic activity; 3) ANP has inhibitory actions on sympathoadrenal activity in vivo.

The direct adrenergic stimulation of regulated atrial ANP release is mediated via activation of β- and αl-adrenoceptors; it interfers with indirect effects of sympathetic activity affecting mechanical stimuli of atrial ANP release (i.e.: diastolic atrial distension and frequency of atrial contraction).

The reflex modulation of sympathetic activity during ANP-induced hypotension and the direct ANP-induced sympathoinhibition counteract each other during exogenous ANP application, but they act in concert during elevations of atrial filling, which releases ANP and concomitantly activates sympathoinhibitory reflexes. ANP can inhibit the sympathoadrenal system by several mechanisms: by activation of mechanosensitive vagal type C afferents eliciting sympathoinhibitory reflexes, by activating ANP binding sites on sympathetic ganglia and on CNS areas outside the bloodbrain barrier, and by inhibiting norepinephrine release from postganglionic nerve terminals and epinephrine release from the adrenal medulla.

The sympathoinhibition by ANP is not attenuated by prolonged elevations of circulating ANP during development of congestive heart failure, when other ANP effects are reduced due to target organ desensitation. Thus, the relative importance of the sympathoinhibition within the broad spectrum of ANP effects is augmented during prolonged ANP exposure.
