Zusammenfassung

Die 1906 von Alois Alzheimer bei einer nach 4 l/2jährigen Krankheitsdauer verstorbenen 55 jährigen Demenzpatientin klinisch und morphologisch beschriebene und nach ihm benannte Krankheit gilt heute als eines der größten medizinischen und gesundheitspolitischen Probleme der zivilisierten Welt. Sie ist die häufigste Ursache von Demenzen im höheren Lebensalter: in Autopsieserien stellt sie 45–80% neben oder in Verbindung mit vaskulärer Demenz (je 10–20%) bzw. anderen Prozessen in 10–15% [22]. Die praktische Bedeutung der Alzheimer-Krankheit (AD) zeigen die allein in USA für die institutionelle Behandlung von AD-Patienten geschätzten Kosten von 25 Mrd. Dollar pro Jahr [26] und die von der US-Regierung für die Fiskaljahre 1988-90 für AD-Forschung genehmigten Mittel in Höhe von 1,157 Mrd. Dollar (US CARE Act HR 3130-87). Trotz einer wohl nur von der AIDS-Forschung übertroffenen wissenschaftlichen Aktivität sind Ursachen, spezifische Laborbefunde (Marker) zur Frühdiagnostik der AD, ihre Beziehung zum phiologischen Altern sowie wirksame Strategien zur Behandlung und Vorbeugung der AD bisher unbekannt.
