Abstract

The thylakoid membrane of plant chloroplasts is the site of the primary steps in the photosynthetic conversion of light to chemically bound energy. This is a complicated process that requires several partial reactions, including light capture, charge separation, electron transport, proton translocation, and enzyme catalysis. The coordination and regulation of this process require a defined molecular organization of the thylakoid membrane. A great deal of our present understanding of the photosynthetic process has been made possible by the ability to perform structural and functional studies on isolated thylakoids, subthylakoid fractions and pure supramolecular protein complexes. The aim of this chapter is to give a methodological description of the main isolation procedures available for such thylakoid material.
