Phylogenesis of the Vertebrate Immune System

Abstract

Our current knowledge of the phylogenesis of immunity is exclusively based on studies of comparative immunity. We know, for example, that some functional components of the vertebrate immune system such as allogeneic recognition and allograft rejection are found in living representatives of primitive diploblastic invertebrates, whereas others, such as specific immunological memory, appear only in more advanced invertebrates where, as in vertebrates, they are associated with the presence of lymphocyte-like cells (reviewed in Ref. 43). We also know that the vector of antibody activity, the multichain immunoglobulin (Ig) molecule, has been detected so far in vertebrates only (reviewed in 61). If these comparative studies have provided a considerable harvest of facts and a reasonable picture of the possible evolution of the immunse system, they have also left us with the disquieting feeling that they may not deal with evolution itself but rather with the present immunological status of living highly evolved representatives of extinct species. This is why, after reviewing the expression of some elements of the mammalian immune system in a variety of primitive species (Ig molecules, lymphocyte diversity, and major histocompatibility complex), it seemed worthwhile to present a new approach to the study of the evolution of the immune system. This approach was made possible by the existence of species where the role of gene duplication in evolution (63) can be more or less mimicked via the production of “artificial” polyploid individuals. An example will be given in Xenopus, the South African frog, where the genetic control of acute graft rejection and the mixed leukocyte reaction in “artificially” produced polyploid individuals and an naturally occurring polyploid species have been compared.
