T4-Induced Modification of Escherichia coli RNA Polymerase in vitro

Abstract

This paper reports an in vitro system derived from T4-infected E. coli B which will modify the α polypeptides of purified E. coli RNA polymerase. The system allows direct investigation of the chemical structure introduced by modification and could provide an assay for purification of the enzyme(s) responsible. Results to date with this in vitro system, plus chemical studies on in vivo-modified α polypeptide, suggest that adenosine diphosphate ribose donated from NAD+ is linked through its terminal ribose to the guanido nitrogen of a unique arginine in the α polypeptide.

Most of this work was carried out at Harvard University, supported by NIH grant CM-09541, while the author was an NSF Predoctoral Fellow.
