Cloning and Analysis of an Azospirillum brasilense Iteron and hslUV Operon Containing Region

Abstract

A 3 kb DNA region from Azospirillum brasilense strain SPF94, located upstream of the histidine biosynthetic cluster, was cloned and analysed. The 5′ region of this sequence contained two putative ORF sharing a very high degree of sequence homology with the Escherichia coli hslUV heat-shock operon.

The region between the A. brasilense hslUV genes and the histidine operon did not contain any ORF but the analysis performed by mutual informational algorithm permitted to detected 8 tandemly repeated sequences of 21 bp. Since iterons are known to act as protein binding sites, we suppose that this sequence could play a role in regulating the histidine or thehslUV operons. Moreover the computational analysis of the DNA topology has revealed that this region is remarkably curved, a feature generally associated to protein binding sites.
