Zusammenfassung
Gastrointestinale Hormone und Zytokine beeinflussen in komplexer Weise das exokrine Pankreas synergistisch und antagonistisch. Exemplarisch werden die 3 Bereiche Sekretion, Wachstum und Genregulation an einigen Beispielen erläutert. Die Sekretion pankreatischer Enzyme wird durch gastrointestinale Hormone/ Neuropeptide über G-Protein-gekoppelte Rezeptoren reguliert, die als Signaltransduktionswege im wesentlichen cAMP oder IP3/Ca2+/DAG benutzen. Daneben wurde in jüngster Zeit die Rolle von Tyrosinkinasen bei der Regulation der Sekretion untersucht. Hiermit ergibt sich eine direkte Interaktionsmöglichkeit von Hormonen und Zytokinen an der Azinuszelle. Über endokrine, parakrine und autokrine Interaktion modulieren gastrointestinale Hormone und Zytokine das Wachstum des exokrinen Pankreas. Eine Interaktion kann auf der Ebene der Rezeptorexpression, der Postrezeptor-Signaltransduktionsmechanismen und der Ebene der Genregulation erfolgen. Zytokine und Wachstumsfaktoren scheinen über ihre häufig als Rezeptor-Tyrosinkinasen-fungierenden Rezeptoren eine entscheidende Rolle bei der Unterhaltung autonomen Wachstums zu spielen. Die Genregulation im exokrinen Pankreas wird in vielfältiger Weise von gastrointestinalen Hormonen und Zytokinen moduliert. Die Aktivierung des für Wachstum bedeutsamen Transkriptionsfaktorkomplexes AP1 durch CCK, die in ähnlicher Weise durch EGF erfolgen kann, ist ein Beispiel. Die schnelle Modulation der Expression von Pankreatitis-assoziiertem Protein (PAP) durch Interferon-γ könnte bei der Induktion einer akuten Pankreatitis bedeutsam sein. Die Induktion von AP1 und die Regulation der Expression von PAP eignen sich als Modellsysteme für die Interaktion von gastrointestinalen Hormonen und Zytokinen auf der Ebene der Genregulation im Pankreas.
Summary
Gastrointestinal hormones and cytokines synergistically and antagonistically influence the exocrine pancreas in a complex way, as can be demonstrated in the regulation of secretion, growth and gene expression. Secretion of pancreatic enzymes is regulated by gastrointestinal hormones/neuropeptides via G protein coupled receptors, linked to the cAMP or inositol trisphosphate/calcium/ diacylglycerol signal transduction pathway. Recently, the role of tyrosine kinases in the regulation of secretion has been characterized. This demonstrates a direct interaction of hormones and cytokines on the level of the acinar cell. Gastrointestinal hormones and cytokines influence exocrine pancreatic growth via endocrine, paracrine and autocrine pathways. The interaction has been demonstrated at the level of receptor expression, postreceptor signal transduction pathways and gene expression. Receptor tyrosine kinases for cytokines and growth factors seem to be crucially involved in the process of autonomic growth. Gene expression in the exocrine pancreas is directly and indirectly modulated by gastrointestinal hormones and cytokines. CCK and EGF activate the heterodimeric transcription factor AP1 which is important for the initiation of growth. The expression of pancreatitis-associated protein (PAP) is rapidly induced by interferon-γ, a process that may be relevant during the early phase of acute pancreatitis. Induction of AP1 and expression of PAP represent model systems to study the interation of gastrointestinal hormones and cytokines at the level of gene regulation in the exocrine pancreas.
