Zusammenfassung

Glutamat, Aspartat und weitere Aminosäuren sind die Agonisten exzitatorischer Synapsen des zentralen Nervensystems und werden an ca. 40% aller Synapsen präsynaptisch freigesetzt (Mattson et al. 1993; McDonald u. Johnston 1990; Mikkelsen et al. 1993; Lipton u. Rosenberg 1994). Während extrazelluläre Glutamatkonzentrationen von 0,3–0,6 μM physiologisch sind, liegt die neurotoxische Konzentration mit 2–5 μM nur wenig darüber. Die parenchymatöse, intrazelluläre Glutamatkonzentration ist mit 3–10 mM 10 ooofach höher als die extrazelluläre (Coyle u. Puttfarcken 1993; Lipton u. Rosenberg 1994). Für die Erhaltung dieses Glutamatgra-dienten sind hochaffine, energieabhängige Transportsysteme zuständig.
