Die Rolle von Tissue Factor in der Tumorbiologie

Zusammenfassung

Tissue Factor ist ursprünglich als Gewebsthromboplastin bezeichnet worden. Tissue Factor ist der Faktor-VII-Rezeptor, an den Faktor VII gebunden, die Gerinnungskaskade aktivieren kann. Tissue Factor ist nicht nur ein “immediate early gene”, das bei Zellteilung aktiviert wird, seine Expression ist auch von großer Bedeutung für die Tumorbiologie. Überexpression von Tissue Factor führt zur vemehrten Freisetzung angiogenetisch aktiver Faktoren, v.a. dem “vascular endothelial cell growth factor” (VEGF). Blockade der TissueFactor-Expression führt zur verminderten Angiogenese durch vermehrte Freisetzung antiangiogenetischer Faktoren. Tissue Factor ist damit nicht nur ein zentraler Regulator der Gerinnung, sondern auch der Angiogenese solider Malignome.
